(1)药效学:本品具有镇痛、抗炎及解热作用。本品通过抑制环氧酶使组织局部前列腺素的合成减少及抑制白细胞的趋化性和溶酶体酶的释放而起到药理作用。本品治疗关节炎时的镇痛、消肿等疗效与吲哚美辛、阿司匹林、萘普生相似。但由于本品抑制环氧酶-2所需的浓度高于抑制环氧酶-1的浓度，因此胃肠道的不良反应较多。

(2)药动学:口服吸收好。食物可降低吸收速度，但不影响吸收总量。血浆蛋白结合率高达90%以上。经肝脏代谢。半衰期平均为50小时(30~86小时)，肾功能不全患者半衰期延长。由于半衰期较长，一次给药即可维持24小时的血药浓度相对稳定，多次给药易致蓄积。一次服药20mg，3~5小时血药浓度达峰值，血药有效浓度为1.5~2μg/ml。血药稳定浓度在开始治疗后7~12天方能达到。66%自肾脏排泄，33%自粪便排泄，内有<5%为原形物。
